Meaning: No hope is left for somebody; doomed to failure.

もう望みがない、失敗に終わる。

How to use: If they hadn't scored that last second goal, it would have been all up with them.

もし、あの最後の1秒のゴールが決まっていなければ、彼らは何もかもおしまいだったはずだ。

このupは「終わった」という意味で、be 動詞+ all up with +人の形で、「全く望みがない、万事休すである」となります。
